Fullerene derivatives as dual inhibitors of HIV-1 reverse transcriptase and protease

نویسندگان

چکیده

In the present study, we newly synthesized three types of novel fullerene derivatives: pyridinium-type derivatives trans-3a and 4a-5b, piperidinium-type derivative 9, proline-type 10a-12. Among assessed compounds, 5a, 10e, 10f, 10i, 11a-d, 12 were found to inhibit both HIV reverse transcriptase protease (HIV-PR), with IC50 values in low micromolar range being observed. Regarding HIV-PR inhibition activity, 11a-11d 12, bearing an alkyl chain between hydroxylmethylcarbonyl (HMC) moiety pyrrolidine ring, more potent than other derivatives. This result might indicate that connecting HMC moieties through properly sized leads improved inhibitory activity.

Synthesis and molecular docking of novel N-((2-chloroquinolin-3-yl) methylene)-4-methylbenzenamine derivatives as anti-HIV-1 reverse transcriptase inhibitors

In this research work, a proficient method has been developed for the preparation of novel N-((2-chloroquinolin-3-yl) methylene)-4-methylbenzenamine derivatives from 2-chloroquinoline-3-carbaldehyde derivatives and p-toluidine in ethanol as solvent and using catalytic amount of acetic acid under reflux conditions to obtain desired products in good yields. The identification of all the synthesiz...

Design and synthesis of tetrahydrophthalimide derivatives as inhibitors of HIV-1 reverse transcriptase

BACKGROUND Non-nucleoside reverse transcriptase inhibitors (NNRTIs) are one of the key components in highly active anti-retroviral therapy because of their high specificity and less toxicity. NNRTIs inhibit reverse transcriptase enzyme by binding to the allosteric site, which is 10Å away from the active site. Rapid emergence of resistance is the major problem with all anti-HIV agents. Hence, th...

Rationally designed dual inhibitors of HIV reverse transcriptase and integrase.

Bifunctional inhibitors were designed and synthesized based on 1-[(2-hydroxyethoxy)methyl]-6-(phenylthio)thymine (HEPT)a1 non-nucleoside reverse transcriptase (RT) inhibitors and diketoacid (DKA) integrase (IN) inhibitors. Biochemical studies revealed activity against RT and IN at low nanomolar and low micromolar concentrations, respectively. Exceptionally low IC50 values from a cell-based assa...
